چکیده

The present study assessed the in vivo effects of fatty acids on inflammation and carcinogenesis in mice. Fifteen fatty acids were examined for their effects on the inhibitory effect of caffeine on 12-O-tetradecanoylphorbol-13acetate (TPA)-induced ear oedema in mice. Furthermore, linoleic acid was studied for its effects on the inhibitory effect of caffeine on carcinogenesis in mouse skin initiated with 7,12-dimethylbenz[a]anthracene (DMBA) and promoted by TPA. Among fatty acids, linoleic, -linolenic and oleic acids most strongly blocked the inhibitory effect of caffeine on TPAinduced ear oedema. Furthermore, linoleic acid also markedly blocked the inhibitory effect of caffeine on the tumourpromoting activity of TPA. This is the first report to suggest that fatty acids, such as linoleic, -linolenic and oleic acids, block the anti-inflammatory activity of caffeine on TPA-induced inflammation in mice. Linoleic acid blocked the inhibitory effect of caffeine on tumour promotion by TPA in the two-stage mouse skin carcinogenesis model. These results suggest that intake of linoleic acid requires attention.

Inhibitory Effects of Gymnema (Gymnema sylvestre) Leaves on Tumour Promotion in Two-Stage Mouse Skin Carcinogenesis

Ethanol extracts of gymnema (Gymnema sylvestre) leaves exhibited marked antitumour-promoting activity in an in vivo two-stage carcinogenesis test in mice using 7,12-dimethylbenz[a]anthracene as an initiator and 12-O-tetradecanoylphorbol-13-acetate (TPA) as a promoter. From the active fraction of the ethanol extract of the gymnema leaves, three triterpenoids were isolated and identified. These c...

Inhibitory effect of stevioside on tumor promotion by 12-O-tetradecanoylphorbol-13-acetate in two-stage carcinogenesis in mouse skin.

Four steviol (ent-kaurene-type diterpenoid) glycosides, stevioside, rebaudiosides A and C, and dulcoside A, have been isolated from Stevia rebaudiana BERTONI. These compounds showed strong inhibitory activity against 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced inflammation in mice. The 50% inhibitory dose of these compounds for TPA-induced inflammation was 54.1-291.6 micro g/ear. Further...

Linoleic acid metabolite suppresses skin inflammation and tumor promotion in mice: possible roles of programmed cell death 4 induction.

(+/-)-13-Hydroxy-10-oxo-trans-11-octadecenoic acid (13-HOA) is one of the lipoxygenase metabolites of linoleic acid (LA) from corn germ. Recently, we reported that this metabolite suppressed the expression of lipopolysaccharide-induced proinflammatory genes in murine macrophages by disrupting mitogen-activated protein kinases and Akt pathways. In this study, we investigated the inhibitory effec...

Inhibitory Effects of Brazilian Propolis on Tumor Promotion in Two-Stage Mouse Skin Carcinogenesis

Propolis is produced by honeybees and has many biological properties, including immunomodulatory, antiinflammatory, anti-oxidant, anti-bacterial, anti-viral and anti-cancer actions. Five ethanol extracts of Brazilian propolis were tested for inhibition of 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced inflammation in mice. The ethanol extract of Brazilian propolis AF-08 markedly inhibited T...

Differential Effects of Dietary Linoleic Acid on Mouse Skin-Tumor Promotion and Mammary Carcinogenesis1

On the basis of reports of rat mammaryand pancreas-tumor models, we hypothesized that an increase in consumption of linoleic acid (LA) would also cause an enhancement in mouse skin-tumor promotion. SENCAR mice were placed on diets containing 0.8%, 2.2%, 3.5%, 4.5%, 5.6%, 7.0%, or 8.4% LA, 1 week after initiation with 7,12-dimethylbenz(a (anthracene and 3 weeks before starting promotion with 12-...
